PHILADELPHIA, Nov. 3, 2026 /CourierPR/ -- Verismo Therapeutics, a clinical-stage CAR T cell therapy company, has presented new preclinical data at the Society for Immunotherapy of Cancer (SITC) 41st Annual Meeting in Phoenix, Arizona, highlighting the enhanced efficacy and reduced cytokine release of its SynKIR™-110 KIR-CAR T cell therapy compared to conventional CD28-based single-chain CAR T cells.

The data, presented by Dr. Nora Yucel from Verismo Therapeutics, demonstrated that SynKIR™-110 achieved faster tumor control, with approximately 10-fold lower interferon-gamma (IFNγ) and 30-fold lower tumor necrosis factor-alpha (TNFα) and interleukin-2 (IL-2) production. This finding is significant as it suggests a more targeted and less inflammatory response, which could lead to improved patient outcomes.

In vivo preclinical models showed that tumor-infiltrating SynKIR™-110 T cells exhibited reduced exhaustion markers, such as programmed cell death protein 1 (PD1), lymphocyte activation gene 3 (LAG3), and T cell immunoglobulin and mucin domain 3 (TIM3), compared to CD28- and 4-1BB-based single-chain CAR T cells. Additionally, the study found that SynKIR™-110 did not accumulate in healthy lung tissue, a common issue with conventional single-chain CAR T therapies.

The company's KIR-CAR platform, which uses NK cell-derived killer immunoglobulin-like receptor (KIR) and DAP12 to separate antigen binding from T cell activation, has shown promise in delivering a more durable and effective anti-tumor response. This approach is designed to reduce the tonic signaling and non-specific off-target activity associated with conventional single-chain CAR T designs.

Verismo Therapeutics is currently evaluating SynKIR™-110 in the ongoing Phase 1 STAR-101 clinical trial (NCT05568680) for patients with advanced mesothelin-expressing solid tumors, including mesothelioma, ovarian cancer, and cholangiocarcinoma. Initial clinical data from the trial, presented at the American Association for Cancer Research (AACR) 2026 meeting, showed a favorable safety profile with no dose-limiting toxicities, no Grade 3 or higher cytokine release syndrome, and no immune effector cell-associated neurotoxicity syndrome (ICANS) observed. The trial also demonstrated tumor reduction in patients.

SynKIR™-110 is part of Verismo Therapeutics' portfolio of KIR-CAR assets, which also includes SynKIR™-310 (NCT06544265). The company is the only entity developing the KIR-CAR platform, which aims to improve T cell functional persistence and reduce exhaustion, leading to enhanced efficacy against challenging tumors.

The findings presented at SITC 2026 underscore the potential of the KIR-CAR platform to address unmet medical needs in the treatment of advanced solid tumors. Verismo Therapeutics continues to advance its research and clinical trials to bring innovative therapies to patients in need.
